LOAA Digital PCR System
LOAA™ dPCR
is a new generation system combining digital PCR and Real-Time PCR technology.
Most currently available digital PCR systems use the so-called "end-point" analysis, which is similar to the principle of conventional PCR, where information about the result of the amplification can be obtained only at the end of the PCR analysis.
Similarly, in digital PCR instruments based on "end-point" analysis, the fluorescence signal is read only at the end of the reaction, and not in real time during each cycle, which is typical for Real-Time PCR analysis.
In contrast to these systems,
LOAA™ dPCR
is based on digital PCR technology, in which the reaction mixture is distributed by a microfluidic system into
20 000 nanowells
, in each of them a separate PCR reaction takes place, where the signals are read in real time mode. As a result, the system generates an amplification curve for every single reaction of
20 000 nanowells
.
Reading the fluorescence signals in real time mode has a significant advantage, because by visualizing the amplification curves, the user can judge the presence of a non-specific reaction and have additional control over the analytical process.
System capacity, models and configurations
LOAA™ dPCR
is available in two models -
LOAA Analyzer (On-Point)
and
LOAA-M
. The system can be configured differently to provide analysis of one to 16 samples simultaneously.
1. The LOAA Analyzer (On-Point)
is a single-module system for analysis of single samples. The distribution of the master mix, amplification, signal reading and system management are all performed within the same module, which greatly facilitates the workflow, making it easy and intuitive.
2. LOAA-M
is a system that can be configured and upgraded according to individual user needs and workflow. LOAA-M consists of two modules:
LOAA-M Console
- through which the system is operated
LOAA-M Analysis module
– allowing analysis of 1 to 4 samples simultaneously
The LOAA-M
can be upgraded with 1 to 4 separate analysis modules to provide a full analysis capacity of 16 samples.
LOAA™ dPCR
was developed for both in vitro diagnostic and research purposes.
Diagnosis and monitoring of chronic myeloid leukemia
is part of the validated diagnostic solutions for the system, which is performed with the
Dr.PCR™ BCR-ABL1 Major IS Detection Kit
.
LOAA™ dPCR in combination with Dr.PCR™ BCR-ABL1 Major IS Detection Kit is an integrated diagnostic solution for the quantification of BCR-ABL1 transcripts (e13a2, e14a2) in clinical blood samples.
Main features of the
LOAA™ dPCR System
and
Dr.PCR™ BCR-ABL1 Major IS Detection Kit
:
Analysis technology by digital PCR (dPCR)
Simple and intuitive workflow – the sample is aliquoted directly into a chip which is then loaded into the system
Analysis time: 1 hour and 30 minutes
Quantitative analysis of BCR-ABL1 and ABL1 transcripts without the need of using quantitative standards
Integrated barcode reader for scanning and traceability of samples
The system generates an automatic report with the patient result in which the result is expressed as %IS, MR Level, NCN (%) according to international guidelines and standards for reporting BCR-ABL1 results.
Sensitivity of detection - MR 4.6 (0.0025% IS)
Dynamic range - 50 – MR 0.3 - MR 4.6 (0.0025%IS)
